Inhibition of human tumour prostate PC-3 cell growth by cannabinoids R( þ )-Methanandamide and JWH-015: Involvement of CB2

BACKGROUND: We have previously shown that cannabinoids induce growth inhibition and apoptosis in prostate cancer PC-3 cells, which express high levels of cannabinoid receptor types 1 and 2 (CB1 and CB2). In this study, we investigated the role of CB2 receptor in the anti-proliferative action of cannabinoids and the signal transduction triggered by receptor ligation. METHODS: The human prostate cancer cell lines, namely PC-3, DU-145 and LNCaP, were used for this study. In the early stages, prostate cancer is androgendependent; therefore, medical castration has shown significant results during the initial stages of this pathology. Despite this early effect, advanced prostate cancer is resilient to such treatment. The endocannabinoid system and cancer: therapeutic implication

The endocannabinoid system is implicated in a variety of physiological and pathological conditions (inflammation, immunomodulation, analgesia, cancer and others). The main active ingredient of cannabis, D9 -tetrahydrocannabinol (D9 -THC), produces its effects through activation of CB1 and CB2 receptors. CB1 receptors are expressed at high levels in the central nervous system (CNS), whereas CB2 receptors are concentrated predominantly, although not exclusively, in cells of the immune system. Endocannabinoids are endogenous lipid-signalling molecules that are generated in the cell membrane from phospholipid precursors.
